The basic principles for selecting antigen peptides include
(1) Try to be on the protein surface as much as possible;
(2) Ensure that the sequence does not form alpha helix;
(3) The N-terminal peptide segment is better than the middle peptide segment;
(4) Avoid repeating or near repeating sequences within the protein;
(5) Avoid peptides with strong homology;
(6) Crosslinking can be done at both ends of N and C, and the selection criteria are based on crosslinking at the less important end for antibody production;
(7) There should not be too many Pros in the sequence, but having one or two Pros is beneficial as it can make the peptide chain structure relatively stable, which is beneficial for producing specific antibodies.
