Guidelines for ¦Â - amyloid protein aggregation |
|
1. Dissolve the powder of A ¦Â peptide (1-42) in cold hexafluoro-2-propanol (HFIP). Incubate the peptide solution at room temperature for at least 1 hour to establish monomeric and randomized structures.
2. Evaporate HFIP and store the obtained peptide in thin film form at -20 or -80 ¡æ.
3. Dissolve the obtained film in anhydrous DMSO at a concentration of 5 mM, and then dilute it to the appropriate concentration and buffer (culture medium without serum and phenol red) using a vortex method.
4. Next, age the solution at 4-8 ¡æ for 24 or 48 hours. Then centrifuge at 14000g for 10 minutes at 4-8 ¡æ; Soluble oligomers are present in the supernatant. Dilute the supernatant 10-200 times for subsequent experiments.
